Muscle Satellite Cells Cloning and Culture Derived from Hanwoo and Holstein Cattle


Abstract
The objectives of this study were to exam optimum culture condition and to compare growth rate of muscle satellite cells derived from Hanwoo and Holstein cattle. Animal growth depends on the proliferation and differentiation of muscle satellite cells. Muscle satellite cells were separated by pronase digestion from 250g muscle and cloned pure cell lines. The optimum cell culture condition was tested with three different culture medium (DME / F12, DMEM, McCoy¢¥s 5A) and three different source of serums (fetal calf serum, chicken serum, horse serum). The number of nuclei per unit area was lower in DME/F12 (68%) and McCoy¢¥s 5A (64%) than that of DMEM (100%) (P$lt;0.05). Also cell growth rate was lower in CS(37.3%) and HS (10.2 %) than that in FCS (P$lt;0.05). Therefore. the optimum muscle satellite cell culture condition was DMEM + 10 % FCS in both Hanwoo and Holstein cattle.
